LEAP HDX automation
The LEAP HDX platform is an advanced, automated scheduling and experimental workstation providing ease of use, high reproducibility and exceptional data quality for HDX experiments.
Labeling
HDX relies on accurate timing of the labeling step.
Quenching
The system will optimize the timing of the labeling step to ensure quenching is just in time for the LC-MS injection, using a chilled syringe. A typical experiment might consist of a dozen time points, in triplicate, for each protein sample. The LEAP HDX system will schedule the experiments and perform all steps for each time point of every sample. Long labeling experiments are automatically scheduled first to ensure the shortest possible instrument time.
Optimized HDX fluidics
The introduction of optimized HDX fluidics that enables full fluidic path backflushing, extending column life and maintaining consistent retention times to deliver higher quality data.
The new fluidics pathway is built upon UHPLC rated valves and column hardware (21 kpsi), with flow paths minimized to reduce swept volumes.
Trajan manufactured HDX specific pre-cut, high pressure lines and fittings, designed to improve reliability and enhance performance. All fluidic components are designed to offer consistently reproducible results.
ChronosHDX control software
Meeting the challenge of producing the highest quality data sets.
HDX experiments require time-sensitive operation. Therefore, in traditional methods, an operator has to attend throughout the course of HDX experiments. However, time-controlled automation enables unattended operation using intelligent scheduling with Chronos software.
Trajan uses ChronosHDX scheduling software to automate the precise labeling and quenching of samples, as well as injection and digestion of proteins on the LC-MS analysis system.

ProDx consumables are specifically designed and developed for use with the LEAP HDX platform.
The ProDx Protease column is optimized to perform at 0°C, the ideal temperature for performing HDX reactions.
Improved protease coupling reduces carryover from non-specific binding, while lower backpressures minimize the potential for contamination and overpressure leakage.
When used in combination with the new LEAP HDX valve system, it can be backflushed automatically to remove frit-loaded or undigested intact material. Clean columns give consistent retention times, overcoming traditional issues with non-selective binding and poor reproducibility.

Automated Hydrogen-Deuterium Exchange (HDX) experiments using the LEAP HDX platform.
ProDx columns were developed specifically for use with the LEAP HDX system. Packed in bio-inert stainless steel glass lined columns to withstand high pressure digestion practices, making it compatible with other systems.
The ProDx Protease (Pepsin) column is the first in a range of immobilized Protease columns available from Trajan. The initial intended use is with the LEAP HDX workflow, where these columns are used in-line for protein-protein and protein-ligand studies. The columns can be deployed outside of the HDX workflow as appropriate, possible alternate configurations might include general proteomics workflows, protein purification studies and QC applications. Column chemistry is such that limited exposure to common denaturing reagents and quenching salts (such as 6.4M Guanidine Hydrochloride, 8M Urea and 4M TCEP) do not adversely impact proteolytic performance.
ProDx Protease (Pepsin) columns are delivered chilled for optimal performance. Columns need to be maintained between 2-8°C. Column performance can be reduced if the column is maintained and operated above 15°C for extended periods of time, or if exposed to non-acidified aqueous reagents or organic reagents.

ProDx consumables are specifically designed and developed for use with the LEAP HDX platform.
ProDx trap columns are optimized for maximum recovery of peptide fragments, efficient desalting and low backpressures during the digestion and elution processes of HDX runs.
While many columns used for HDX trapping are not designed for use with low temperature HDX systems, ProDx trap columns are designed to operate down to 0°C, with consistent recovery, and demonstrate reduced backpressure at standard conditions of 200 μL/min, 100% Aqueous and 0°C, lowering instances of leakage and need for troubleshooting.
ProDx trap columns are pre-configured with PEEKsil® lines for ease of use, reducing the number of fittings needed in the system, preventing issues that commonly occur when relying on other columns with multiple connections.

Upgrade sample throughput without worrying about carryover
The LEAP PAL injector head is a syringe pump itself, making it the ideal sample delivery system for ESI flow rates, unlike conventional HPLC pumps.
Many lipids are notorious for their adherence to glass and other surfaces resulting in carryover between samples. The special LCMS syringe tool incorporates unique features to minimize this carryover to below 1% for the abundant neutral and phospholipids and even lower for other species.
The illustration below shows how this rapid washing is achieved. After washing the inside, the system will wash the needle externally and be ready to aspirate the next sample.

Greatest experiment control with powerful, user-friendly software
The infusion process is controlled using our Chronos software, allowing maximum compatibility with your preferred Chromatography Data System, LIMS and MS data systems.
Method parameters are available for flow rates, infusion times, stabilization delays and cleanup conditions, MS acquisition method switching.
With an optional second valve, standard chromatography methods can be used in the same sequence with direct infusion samples.
