High-throughput, high sensitivity and low carryover: the new infusion automation from Trajan offers you everything you need to bring your lipidomics mass spec workflow to its peak performance.
Move from manual sample infusion to the highest standard of infusion automation. With rapid cleanup between injections and best-in-class software architecture to integrate into your workflow, our infusion automation will elevate your lipidomics experiments.
The most consistent and flexible system available for infusion automation
Fully customizable infusion eliminates carryover concerns while saving you time. Allows for sample infusion at flow rates between 5 and 100 µL/min — directly into your ESI source via the injection valve — precisely and without pulsation. Compatible with both shotgun and targeted lipid analysis.
The powerful Chronos software can execute important operational and integration functions such as reading and reporting to LIMS software, as well as MS acquisition mode switching within a single infusion. The system maintains flow to the source while quickly and thoroughly washing the syringe system for the next infusion.
Exclusive inert configuration is available for your trickiest lipid classes — ensuring your samples never come into contact with any polymers.
“We just ran a whole pile of samples. It ran 24/7 without a hitch. Very reliable.” - Calvin Vary, PhD, Proteomics and Lipidomics Core Facility Maine Medical Center Research Institute, Maine, USA.
Upgrade sample throughput without worrying about carryover
The LEAP PAL injector head is a syringe pump itself, making it the ideal sample delivery system for ESI flow rates, unlike conventional HPLC pumps.
Many lipids are notorious for their adherence to glass and other surfaces resulting in carryover between samples. The special LCMS syringe tool incorporates unique features to minimize this carryover to below 1% for the abundant neutral and phospholipids and even lower for other species.
The illustration below shows how this rapid washing is achieved. After washing the inside, the system will wash the needle externally and be ready to aspirate the next sample.

Greatest experiment control with powerful, user-friendly software
The infusion process is controlled using our Chronos software, allowing maximum compatibility with your preferred Chromatography Data System, LIMS and MS data systems.
Method parameters are available for flow rates, infusion times, stabilization delays and cleanup conditions, MS acquisition method switching.
With an optional second valve, standard chromatography methods can be used in the same sequence with direct infusion samples.

Since 2002 Trajan has been automating HDX, providing continuous enhancements for rugged HDX sample control and scheduling.
Trajan’s LEAP HDX system is the recognized leader, preferred around the globe by scientists requiring confidence in reproducible results.
Automated HDX provides structural confidence in native states
HDX, especially automated HDX, has uses in development, manufacturing and post-production, providing insight into your protein’s tertiary structure, in its native state(s).
Successful HDX experiments are all about control
With robotic precision, Trajan’s HDX platform exerts exacting control of all key factors:
HDX is a well-established analytical technique for protein-ligand, protein-protein, protein-conjugate interactions and protein stability studies.
HDX uses variations in deuterium exchange rates between more and less accessible/structured regions throughout a protein.
In addition to being faster than other protein techniques like x-ray crystallography and NMR, HDX data can be obtained from samples that are challenging to crystalize and from samples in short supply. It can be used to replicate critical native state conditions, for the most useful results. HDX is an indispensable complementary tool for protein biophysics.
Trajan’s HDX system is compatible with bottom-up, top- and middle-down protein analysis studies.
The total HDX platform
Our total HDX platform covers the complete process: sample input >> the many meticulous steps of preparation >> optimizing mass spec utilization >> clear answers with minimal manual data curation.
Faster answers
Powerful information on protein structure in days – not weeks. Move more quickly, make better decisions.
With HDExaminer, swiftly and easily analyze data from any MS instrument to get a complete picture of the overall deuteration behavior of your protein.
Automation at its finest
The most efficient system control, with unique temperature set points at all stages of the experiment to ensure reliable data. Sustained 0°C environment reduces back exchange, improves sample stability and overall reproducibility.
Our Chronos for HDX software offers a streamlined interface for HDX users and automatically overlaps time points for maximum MS efficiency.
Combines ease of use with advanced options, allowing promotion of user-defined parameters to the sample list, giving the operator control over all aspects of the method without complication.
HDExaminer: a complete analytical tool for HDX experiments
The Trajan LEAP HDX platform works natively with HDExaminer from Sierra Analytics, a complete software solution for HDX analysis, independent of MS data type.
HDExaminer is designed with ease of use in mind, offering numerous ways to visualize your data. These include:
Unique fluidics automation for the best performance
Trajan’s LEAP HDX platform incorporates an advanced cooling chamber for the trapping and chromatography phases which includes an enclosed valve and column chiller as well as solvent pre-conditioning.
Digestion temperature is controlled independently with the protease column chamber, to allow improved digestion efficiency.
A special 3 valve configuration enables zero-downtime backflushing for retention time stability, extended column life and superior carryover protection.
Optimized fittings and precut tubing ensure maximum reliability and instrument up-time.

View the range of ProDx columns
ProDx consumables are specifically designed and developed for use with the LEAP HDX platform.
The ProDx Protease column is optimized to perform at 0°C, the ideal temperature for performing HDX reactions.
Improved protease coupling reduces carryover from non-specific binding, while lower backpressures minimize the potential for contamination and overpressure leakage.
When used in combination with the new LEAP HDX valve system, it can be backflushed automatically to remove frit-loaded or undigested intact material. Clean columns give consistent retention times, overcoming traditional issues with non-selective binding and poor reproducibility.
Automated Hydrogen-Deuterium Exchange (HDX) experiments using the LEAP HDX platform.
ProDx columns were developed specifically for use with the LEAP HDX system. Packed in bio-inert stainless steel glass lined columns to withstand high pressure digestion practices, making it compatible with other systems.
The ProDx Protease (Pepsin) column is the first in a range of immobilized Protease columns available from Trajan. The initial intended use is with the LEAP HDX workflow, where these columns are used in-line for protein-protein and protein-ligand studies. The columns can be deployed outside of the HDX workflow as appropriate, possible alternate configurations might include general proteomics workflows, protein purification studies and QC applications. Column chemistry is such that limited exposure to common denaturing reagents and quenching salts (such as 6.4M Guanidine Hydrochloride, 8M Urea and 4M TCEP) do not adversely impact proteolytic performance.
ProDx Protease (Pepsin) columns are delivered chilled for optimal performance. Columns need to be maintained between 2-8°C. Column performance can be reduced if the column is maintained and operated above 15°C for extended periods of time, or if exposed to non-acidified aqueous reagents or organic reagents.

Unique dynamic flow reservoir
The precision engineered and long lasting dynamic flow reservoir (DFR) allows the LEAP MFx collector to continue collecting chromatographic flow while the automation is moving between wells, resulting in zero-loss.The design achieves a turbulence-free, fully optimized flowpath for collection without peak dispersion.
Undisturbed flowpath for minimal peak diffusion
While flow to the buffer region remains constant during both well filling and robot movement, the buffer region will expand as the DFR travels between wells. This retracts any hanging droplets, and avoids peak diffusion by allowing continuous flow. The buffer region contracts to resume dispensing at the next well.
Setup for collection has never been easier
Tabular and visual method creation
Maintains highest peak resolution with sub-2 second fractions, while providing simplified data correlation, and advanced sample tracking
